Function in PMv: evidence, prediction, innovation
- Gene function
- Prlhr encodes GPR10, the sole high-affinity Gq/Gi-coupled receptor for prolactin-releasing peptide (PrRP, from Prlh), mobilizing intracellular Ca2+. Central signalling curbs food intake and raises energy expenditure; Prlhr-null mice are obese via reduced expenditure, and GPR10 relays leptin-driven thermogenesis from DMH PrRP neurons.
- Region function
- The ventral premammillary nucleus is a small glutamatergic, LepRb-rich nucleus linking metabolic and prolactin status to GnRH/LH secretion, puberty and odour-evoked behaviour.
- Published in this region
- inferred No region-specific literature found. Closest: Roland et al. 1999's ISH map put GPR10 mRNA in reticular thalamus, periventricular and dorsomedial hypothalamus, NTS and area postrema but not premammillary; PrRP-GPR10 function is dissected only in DMH, PVN/SON and brainstem (Gu et al. 2004; Dodd et al. 2014), PMv being defined by LepRb and PACAP neurons (Donato et al. 2011; Ross et al. 2018).
- Predicted function
- Prediction: GPR10 on PMv LepRb glutamatergic neurons is a second peptidergic metabolic input converging with leptin, depolarizing them via Gq/PLC to reinforce drive to GnRH neurons. PMv-restricted Prlhr deletion should delay puberty and blunt the fasting-refeeding LH rebound without changing body weight.
- Innovation
- ★★★★★ 5/5 GPR10 pharmacology and PMv reproductive physiology are mature, yet this receptor has never been examined in this nucleus.
- Tools
- No validated Prlhr-Cre; Prlh-Cre and Prlh/Prlhr knockouts cover ligand and receptor. Brain-penetrant PrRP-31, PrRP-20 and lipidated PrRP31 analogues and GPR10 antibodies exist; PMv is targeted with Lepr-Cre, PACAP-Cre, Vglut2-Cre.
